???metadata.dc.description.resumo???: O fracionamento cromatográfico dos extratos de casca e raiz de Piptadenia gonoacantha J.F. Macbr., de folhas e sementes de Piptadenia rigida Brenan (Leguminosae) e análise das frações através de técnicas cromatográficas e espectroscópicas conduziu ao isolamento e a identificação de constituintes de diferentes classes de metabólitos. Da casca do caule de P. gonoacantha foram isolados cicloartenona, cicloartan-25-26-en-3-ona, 24-metilenocicloartanona, friedelina, 24-metilenocicloartanol, campesterol, sitosterol, estigmasterol, lupeona, lupeol; de raiz desta planta foram isolados cicloartenol, lupeol, campesterol, sitosterol, estigmasterol, uma mistura de isoflavonas, 7-hidroxi-5,8-dimetoxi-isoflavona, 7-hidroxi-3’,4’-metilenodioxiisoflavona e 7-hidroxi-5,8,4’-trimetoxi-isoflavona, além de duas frações ricas em ésteres metílicos. Dos extratos de folhas de P. rigida foram isolados dois poliprenóides, (6E,10E,14E,18E,22E)-2,6,10,14,18,22-hexametiltetracosa-2,6,10,14,18,22-hexeno e (2E,6E,10E,14E,18E)-3,7,11,15,19,23-hexametiltetracosa-2,6,10,14,18,22-hexen-1-ol, fitol e o monoterpeno loliolídeo; de sementes de P. rigida foram identificados um triacilglicerol, uma mistura de ácidos graxos, além do alcalóide indólico bufotenina e o carboidrato pinitol. As estruturas foram determinadas através da análise de dados fornecidos por espectrometria na região do infravermelho, RMN 1H e 13C (técnicas 1D e 2D), de massas incluindo CG-EM e CL-EM-ESI das substâncias naturais. Além da extração de óleos de folhas de P.rigida e P.gonoacantha usando hidrodestilação, utilizou-se fluido supercrítico com CO2 como solvente. Foram testadas diferentes condições operacionais, variando-se principalmente a temperatura (40 a 80 ºC) e a pressão (100 a 200 bar), de modo a buscar os melhores resultados em termos de rendimento e composição do óleo extraído. Os melhores rendimentos na extração de óleo de folhas destas plantas foram de 0,73% a 60 °C e 200 bar, e 1,31% a 80 ºC e 200 bar, respectivamente. A análise dos tempos de retenção, espectros de massas e análise do espectro de RMN 1H permitiu identificar a presença de vários constituintes; tendo como constituintes majoritários presentes no óleo de P.rigida N-butilbenzeno- sulfonamida (em diferentes condições de extração) e seu derivado, além dos esteróides sitostenona, sitosterol, estigmasterol, 4,22-estigmastadien-3-ona, dos triterpenos lupeol, friedelliana, dentre outros. Já no óleo de P. gonoacantha o principal constituinte foi N-butilbenzeno- sulfonamida (a 40º e 100 bar), apesar da semelhança do espectro de RMN 1H, os esteroides e triterpenos foram extraídos em diferentes condições. Realizaram-se análise de teor de fenóis totais e taninos além de propriedades biológicas como antibacteriana e antifúngica de extratos de P. rigida e P. gonoacantha. Desta análise verificou-se que Staphylococcus aureus foi mais sensível ao extrato PGRMA (fração em acetato do extrato metanólico de raízes de P. gonoacantha) rica em fenóis totais e taninos. Como atividade adicional, preparou-se um novo derivado da quercetina tendo um grupo fenil-2,6-dinitro-4-trifluorometila usando a rutina como material de partida. Os resultados da atividade antifúngica deste derivado contra C. albicans foram bastante significativos.
Abstract: The chromatographic fractionation of bark and roots extracts of Piptadenia gonoacantha J.F. Macbr., and from the leaves and seeds of Piptadenia rigida Brenan (Leguminosae) besides the fractions analysis by chromatographic and spectroscopic techniques allowed to the isolation and identification of many constituents belonging to different metabolite classes. From the bark of P. gonoacantha cicloartenone, cicloartan-25- 26-en-3-one, 24-metilenecicloartanone, friedelin, 24-metilenecicloartanol, campesterol, sitosterol, estigmasterol, lupeone, and lupeol were isolated; from the roots of this plant the cicloartenol, lupeol, campesterol, sitosterol, stigmasterol, a mixture of isoflavones, 7- hydroxy-5,8-dimethoxy-isoflavone, 7-hydroxy-3’,4’-metilenedioxy-isoflavone and 7- hydroxy-5,8,4’-trimethoxy-isoflavone were identified, besides two fractions rich in methyl esters. From the leaves extracts of P. rigida two poliprenoids, (6E,10E,14E,18E,22E)- 2,6,10,14,18,22-hexamethyltetracos-2,6,10,14,18,22-hexene and (2E,6E,10E,14E,18E)- 3,7,11,15,19,23-hexamethyltetracos-2,6,10,14,18,22-hexen-1-ol, phytol, and the monoterpene known as loliolid; from the seeds of P. rigida a triacilglicerol, a mixture of fat acids, besides the indolic alkaloid bufotenine and carbohydrate pinitol. The structures were proposed on the bases of IR, 1H and 13C NMR, GC-MS and LC-MS espectrometric analysis and comparison with literature data. Besides the extraction of oils from the leaves of P.rigida e P.gonoacantha by hidrodestillation, and supercritical fluid with CO2 as solvent were used. Different operational conditions were tested, varying the temperature (40, 60 and 80 ºC) and pressure (100, 150 and 200 bar), to find the best yields in the extracted oil composition. The best rendiment in this extraction were 0,73% at 60 °C/200 bar, and 1,31% at 80 ºC/200 bar, respectively, for the both plants, P. rigida and P. gonoacantha. The retection time, mass and 1H NMR spectra analysis, allowed to identify various constituents. The principal constituents in the P.rigida oil were N-butil-benzeno-sulfonamida (at different extraction conditions) its derivatives, besides some steroids such as sitostenone, sitosterol, estigmasterol, 4,22-estigmastadien-3-one, the triterpenes lupeol, friedelin, together other ones. In the P. gonoacantha oil, the principal constituent was N-butil-benzeno-sulfonamida (at 40º/100 bar), despite the similarity between the 1H NMR of the both plant oils the steroids and triterpenes were extracted in different conditions. The total phenols and tannins, besides biological activities such as antibacterial and antifungal activities of the P. rigida and P. gonoacantha extracts were evaluated. In these analysis was observed that Staphylococcus aureus was more sensible to the PGRMA (Ethyl acetate Fraction from the methanolic extract from the roots of P. gonoacantha), that is rich in total phenols and tannins. As an additional activity, a new quercetin derivative with phenil-2,6-dinitro,4- trifluormetil was prepared using the rutin as starting reagent material. The antifungal activity against C. albicans was more significant.
