dc.description.resumoDescreve-se um surto de doença do armazenamento lisossomal em ovinos induzida pelo consumo de Sida planicaulis no Estado do Rio de Janeiro. A ingestão de S. planicaulis Cav. (S. carpinifolia) é responsável pelo aparecimento desta enfermidade em bovinos, equinos, caprinos, ovinos e cervídeos cativos que ocorre, de forma natural, quando a planta predomina na pastagem e há baixa oferta de outras forragens. O principal composto tóxico desta planta, a swainsonina, inibe atividade enzimática de α-manosidase I e II que redunda no armazenamento de glicoproteínas no interior de lisossomos. Este acúmulo de substâncias promove compressão, deficiência no transporte intracitoplasmático e finalmente degeneração em diversos tecidos, dentre os quais o mais afetado é o nervoso. O exame físico dos animais afetados revelou déficit proprioceptivo, incoordenação motora, cambaleio à movimentação e tremores de intenção e na cabeça. Houve aumento acentuado dos tremores em cabeça e membros, com posterior dificuldade de se manter em estação e queda ao solo após a realização do “Head-Raising Test”. A histopatologia evidenciou severa distensão de neurônios de Purkinje, com aspecto espumoso e eventual cariólise ou cariopicnose e intensa vacuolização de células acinares do pâncreas e foliculares da tireoide. Adicionalmente, foram observadas vacuolizações em grandes corpos de neurônios com dissolução do citoplasma associada à marcada proliferação de astrócitos morfologicamente atípicos. Notou-se também múltiplos esferoides axonais de variados tamanhos. O exame lectino-histoquímico positivo para as lectinas Con A, WGA e sWGA foi capaz de caracterizar a enfermidade como uma glicoproteinose. A avaliação ultraestrutural evidenciou numerosos vacúolos de até 2,5μm de diâmetro, delimitados por membranas de até 20nm de espessura em células acinares do pâncreas. O diagnóstico da intoxicação por S. planicaulis foi estabelecido através dos dados epidemiológicos, achados clínico-patológicos e confirmados pelas avaliações lectino-histoquímica e ultraestrutural. A intoxicação natural por S. planicaulis em ovinos ainda não havia sido descrita no estado do Rio de Janeiro.por
dc.description.abstractThis paper describes an outbreak of lysosomal storage disease in sheep induced by Sida planicaulis in the state of Rio de Janeiro. The consumption of S. planicaulis is responsible for this disease in cattle, horses, goat, sheep and captive deer, which occurs naturally, when the plant is abundant and there is shortage of other forage. The main toxic compound in this plant, Swainsonine, inhibits α-manosidasis I e II enzymatic activitiy and causes glycoprotein storage in lysosomes. The storage provokes compression, deficiency in intracytoplasmatic transport, and finally, degenerative changes in various tissues, in which, nervous system is the most affected. Physical examination of the affected animals revealed proprioceptive déficit, motor incoordination, staggers and, neck and head intention tremors. ‘Head-Raising Test’ showed marked increase of head and limb tremors, with sequential difficulty in balance and tumble. Histopathology revealed severe distension of Purkinje cells, with foamy appearance, which eventually progressed to karyolysis and pyknosis, and intense vacuolization of pancreatic acinar and thyroid follicular cells. Additionally, there was vacuolization in neuronal bodies with dissolution of cytoplasm associated with marked proliferation of morphologically atypical astrocytes. Multiple axonal spheroids of variable sizes were noted. Positive reaction in lectin-histochemistry for lectins Con A, WGA, and sWGA was able to characterize the disease as a glycoproteinosis. Ultrastructural evaluation of pancreas sections evidenced numerous vacuoles up to 2,5μm in diameter, circled by 20nm wide membranes in acinar cells. In the present report, diagnosis of S. planicaulis toxicosis was established by epidemiological data, clinical and pathological findings, and confirmed by lectin-histochemistry and ultrastructural evaluation. Natural toxicosis in sheep by Sida planicaulis has not yet been described in the state of Rio de Janeiro.eng
